ABSTRACT

ObjectiveTo investigate the clinical effect of surgery in the treatment of calcified hepatic cystic echinococcosis. MethodsA retrospective analysis was performed for the clinical data of 16 patients with 20 calcified hepatic hydatid cysts who underwent surgical treatment (total pericystectomy, subtotal pericystectomy, and endocystectomy) in The First Affiliated Hospital of Shihezi University School of Medicine from November 2015 to February 2019. ResultsOf all 16 patients, 1 underwent total pericystectomy, 5 underwent endocystectomy, and 10 underwent subtotal pericystectomy. One patient experienced bile leakage after surgery since the cyst ruptured and entered the bile duct and there was a biliary fistula in the outer wall of the cyst before surgery, and the other patients had no residual cavity complications including residual cavity effusion and infection, bile leakage, or jaundice. There was no death and recurrence after operation. ConclusionSurgical treatment is necessary for non-stationary calcified hepatic cystic echinococcosis. Total pericystectomy is not suitable, while subtotal pericystectomy may be the preferred surgical method, especially when the calcified hepatic hydatid cyst is located near the porta hepatis or the intrahepatic and extrahepatic vessels and there is no obvious lacunae between the adventitia and the outer cyst. Endocystectomy can be selected when hepatic hydatid cyst ruptures.

ABSTRACT

BACKGROUND: After the treatment of osteoporotic vertebral compression fractures with bone cement implantation, deteriorative osteoporosis and the distance augment between bone cement and bones result in a decrease in bone cement-bone interface compression strength and torsion strength, and then the bone cement tends to displace, which is easy to cause a secondary vertebral fracture.OBJECTIVE: To investigate the clinical efficacy of bone cement implantation via three approaches on osteoporotic vertebral compression fractures.METHODS: Sixty patients with single osteoporotic vertebral compression fracture were treated with vertebroplasty, percutaneous kyphoplasty or fraction cement vertebroplasty. All data were collected to analyze the correlation between the bone cement morphology and treatment method, and to detect the visual analogue scale, Oswestry disability index,Cobb angle and incidence of secondary vertebral fractures.RESULTS AND CONCLUSION: The visual analogue scale scores, Oswestry disability index, and Cobb angle in thethree groups were significantly improved after treatment (P < 0.05). All above indicators showed no significant difference among groups at each time point after treatment, suggesting that these three kinds of treatments exert the same effects on pain relief, functional recovery, preventing the vertebral height loss and maintaining spinal mechanical properties.Bone cement displacement and secondary vertebral fracture occurred after clotted cement implantation. Therefore, mixed and trabecular-shaped grafts are preferred, aiming to achieve good long-term treatment outcomes in osteoporotic vertebral compression fractures.

ABSTRACT

Objective To observe the morphological changes of pancreatic tissue of thoracic duct ligated rats in fine and ultrastructural levels, and to determine whether lymph block animal model can affect pancreatic islet amyloid polypeptide(PIAP)deposit in rat pancreas. Methods At the 6th month after the operation, some pancreatic tissue sections of 16-month-old experimental rats were embedded in paraffin wax and stained with HE and Congo red;immunohistochemical staining was performed on some frozen sections, which were then observed with light microscope;transmission electron microscope (TEM) specimen preparation and observation were performed on other samples. Results HE and Congo red stained sections showed that the pancreatic glandular lobule space was widened, with significant connective tissue hyperplasia, and fat accumulation when the islet was stained indistinctly or vermeil and tissue space was broadened. The sections with immunohistochemical staining displayed the pancreatic islet as well as the tissues around it were stained into dark brown being positive with PIAP antigen. TEM observation showed the pancreatic glandular interlobule space was widened, while blood vessels and enlarged lymphatic vessels were visible;within widened pancreatic islet interstitial space, a great quantity of lipid droplets and some collagen fibril structures could be seen. Conclusion The ligation of thoracic duct can contribute to pancreatic lymph draining block, lymphagiectasis, connective tissue space and interstitial space widening, fat accumulation, and PIAP deposit in rat pancreas. These structural changes may affect the function of pancreatic islets.

ABSTRACT

Objective To study the expressions of caudal type homeodomain transcription factors CDX1 and CDX2 in different subtype of intestinal metaplasia (IM) and gastric eareinoma, and investigate their roles in the development and progression of IM and gastric carcinogenesis, and determine the relationship between IM and gastric carcinoma. Methods Forty eases of chronic superficial gastritis, 40 cases of chronic atrophic gastritis (CAG), 46 eases of CAG with IM, 40 cases of gastric carcinoma, and 32 eases of IM foci in para-eancerous tissues were selected respectively to construct tissue microarrays. Immunohistechemistry and in situ hybridization were used to assess the expressions of CDX2 protein and CDX1, CDX2 mRNA in different gastric lesions respectively. Results The proportion of type Ⅲ IM in IM foci in para-caucerous tissues was significantly higher than that in CAG (56.25% vs 21.74% ,P< 0.01). There was no expression of CDX1, CDX2 mRNA and CDX2 protein in chronic superficial gastritis and CAG without IM. The expression of CDX2 protein and CDX2 mRNA in CAG with IM, para-cancerous tissues and gastric carcinoma was 69.57%, 53.12%,42.50% (CDX2 protein) and 63.04%,46.88%,35.00% (CDX2 mRNA), respectively. The exprossions of CDX1 mRNA in above throe gastric lesions were 67.39%, 50.00%, 40.00%, respectively. The expressions of CDX2 protein and CDX2, CDX1 mRNA in gastric carcinoma were significantly lower than those in CAG with IM (P< 0.01). But there was no significant difference between gastric carcinoma and IM foci in para-cancerous tissues (P> 0.05). Furthermoro, the expressions of CDX2 protein were significantly associated with histological type of gastric carcinoma, which in intestinal-type was significantly higher than those in diffuse-type(54.55% vs 27.78%, P< 0.05). The expression of CDX2 protein in type Ⅲ IM was also significantly lower than that in type Ⅰ IM(46.43% vs 79.31%,P< 0.05). Conclusions CDX1 and CDX2 may play important roles in the development and progression of IM and gastric carcinoma, and may be new gastric carcinoma associated genes. Type Ⅲ IM is a precancerous lesion of the intestinal-type gastric carcinoma.

ABSTRACT

A new and effective method to produce transgenic animals was established. Without a surgical incision, the recombinant plasmid containing green fluorescence protein (GFP) cDNA was repeatedly injected into male mouse testis at multi-sites. After few weeks of the final injection, the injected male was mated with normal oestrus female to produce transgenic mice. The presence of the GFP cDNA in F1 transgenic individuals were detected by polymerase chain reaction and Southern blot hybridization, which showed that the transgenic rate of mouse F1 offspring was 41%. The transferred gene was integrated into the host genome and could be transmitted to its offspring. When the positive F1 individuals were mated with the wild type ICR mice, the F2 individuals had a transgenic rate of 37%. The results indicate that the high efficiency of gene transfer and the limited number of manipulations make the method suitable for creating a large number of transgenic animals, especially, for producing domestic animals.
